Nonetheless, try guitar > amp, then try, guitar > pedal 1 > amp and so on. if it happens when you plugged straight into your amp then it's your guitar or your amp. You may test out your 1/4 inch output on your guitar.

Alright, let me pose this question. The readings on four different tubes are 23/30, 50/32, 33/52 and 50/35. With the type of tester that you have, what numbers would be relevant to the strength of the tube circuitry and longevity of the tube?
I'm not seeing how that would damage the JCM 800. You sent your JCM into the Fender Effect Loop input. Are you sure that's the only circuit that was made with your amp? I could see potential damage if you had each amps output fighting each other, but the only damage I could imagine happening...
